As an unexpected spin-off from generating the transporter library, we have developed a novel molecular cloning strategy that is simple, fast, flexible, highly efficient, and inexpensive. The method is based on the largely unrecognized, ligase-independent uracil-excision cloning technique, and has great potential for both individual and high-throughput cloning experiments. We foresee great advantages for molecular biologists in using this cloning strategy.
